Expression and functional changes of NOD2 in tongue squamous cell carcinoma

[Abstract] PURPOSE: This study aimed to investigate the expression and functional changes of NOD2 in tongue squamous cell carcinoma(TSCC). METHODS: Immunohistochemical staining and RT-qPCR were employed to evaluate NOD2 expression in TSCC and non-cancer tongue epithelial tissues. hBD-2 production was investigated by immunofluorescence staining and RT-qPCR. Student’s t test was used to assess the statistical significance of mRNA expression between the two groups, and two-way analysis of variance was used to determine the differences among different groups using SPSS 13.0 software package. RESULTS: ①NOD2 was expressed in the cell plasma. NOD2 expression was significantly lower in TSCC tissues (64.0%, 16/25) than in non-cancer tongue epithelial tissues (84.0%, 21/25) (P<0.05). NOD2 messenger RNA expression in TSCC tissues was (0.521±0.080) times of that in non-cancer tongue epithelial tissues(P<0.05). ②hBD-2 was expressed in the cell plasma. When MDP was added to Tca8113 cells for 6,12 h, hBD-2 expression was significantly upregulated (1.770±1.202 and 68.277±48.946 times, P<0.05). CONCLUSION: NOD2 expression was significantly lower in TSCC, hBD-2 expression was markedly upregulated when NOD2 was activated by MDP. This suggests that NOD2 adjust hBD-2 expression in the tongue mucosa, and affect the immune function of TSCC, as well as the progression of TSCC.
